Pathomechanisms of Prenatally Programmed Adult Diseases

چکیده

Based on epidemiological observations Barker et al. put forward the hypothesis/concept that an adverse intrauterine environment (involving insufficient nutrient supply, chronic hypoxia, stress, and toxic substances) is important risk factor for development of diseases later in life. The fetus responds to unfavorable with adaptive reactions, which ensure survival short run, but at expense initiating pathological processes leading adult diseases. In this review, major mechanisms (including telomere dysfunction, epigenetic modifications, cardiovascular–renal–endocrine–metabolic reactions) will be outlined, a particular emphasis role oxidative stress fetal origin

Prenatally programmed hypertension: role of maternal diabetes.

Epidemiological and experimental studies have led to the hypothesis of the fetal origin of adult diseases, suggesting that some adult diseases might be determined before birth by altered fetal development. Maternal diabetes subjects the fetus to an adverse environment that has been demonstrated to result in metabolic, cardiovascular and renal impairment in the offspring. The growing amount of o...

Upregulation of renal BSC1 and TSC in prenatally programmed hypertension.

Prenatal factors, especially intrauterine growth retardation, have been shown to correlate with the risk of essential hypertension in adult life, but the mechanisms are unknown. An experimental model of prenatal programming of hypertension in the rat, induced by a maternal low-protein diet during pregnancy, was employed to study the role of renal Na reabsorption in the pathogenesis. The abundan...

Kidney angiotensin and angiotensin receptor expression in prenatally programmed hypertension.

Adult hypertension may be programmed by the prenatal environment in humans and in experimental animals. The potential role of the intrarenal renin-angiotensin system (RAS) in prenatally programmed hypertension was investigated. Hypertension in rat offspring was induced by maternal protein restriction during pregnancy. The offspring were studied on day 1 of life and immediately preceding the dev...

Castration fails to prevent prenatally programmed hypertension in male rats.

Male offspring of rats that were modestly protein restricted during pregnancy become hypertensive as adults, whereas their female littermates remain normotensive. The purpose of this study was to determine the role of testosterone in promoting this sexual dimorphism of prenatally programmed hypertension. Rats were fed either a normal (19% protein, NP) or modestly protein-restricted (8.5% protei...
